The advantage of playing three defensive players (Linebacker, Lineman, Safety) is that you have more players to play with so it gives you more players to draft or pick up off waivers.  It’s more fun as it’s basically adding three more positions so there’s more higher quality free agents.  Playing with one defensive team is for those who are not into football and just want to stream a defense every week.  It’s not nearly as fun as playing with a team starring Aaron Donald, TJ Watt and Jamal Adams!

(also we can add a defensive player as a keeper next year.  So if you really love Bobby Wagner then you can keep him 🙂

In keeper leagues, the goal is to play like a real NFL general manager!  So it makes it more fun and realistic when you have more players to keep every season.  In real life, NFL teams can’t just draft a brand new team each year.  The more keeper spots, the more chances you can take.  You never know when a player on your team will blow up the following year.  With keeper league, you get to gamble and have more fun!  It also facilitates more off-season trading.

(To those who think they don’t have enough quality players, don’t forget, you can trade draft picks to other teams to acquire players they wish to deal. So every team has the chance to keep 8 or 9 quality players by using the off-season to make trades before the draft.)
